How to reconstitute
  1. Allow both vials (peptide and bacteriostatic water) to reach room temperature.
  2. Wipe the rubber stopper of each vial with an alcohol swab.
  3. Draw 2.00 mL of bacteriostatic water into a reconstitution syringe (use a 3 mL syringe for volumes up to 3 mL).
  4. Insert the needle into the peptide vial at an angle and release the plunger slowly. Let the water run down the side of the vial to avoid foaming.
  5. Gently swirl (do not shake) to dissolve. Most peptides dissolve fully within a minute.
  6. Label the vial with peptide name, concentration (2.50mg/mL), and today's date. Use the Print Label button above for a ready-made label.
  7. Store reconstituted peptide refrigerated at 2-8 degrees C.

The math, the method, and the common questions behind peptide reconstitution.

Reconstitution is the process of adding a solvent (typically bacteriostatic water) to a lyophilised peptide vial to dissolve the powder into an injectable solution. The amount of water you add determines the concentration of the final solution and how many units you need to draw from a syringe to deliver a given dose.

A U-100 insulin syringe is calibrated so that 100 units equals 1 millilitre of fluid. The markings on the barrel run from 0 at the needle end to the maximum volume at the plunger end. A 1 mL syringe shows 0-100 units, a 0.5 mL shows 0-50 units, and a 0.3 mL shows 0-30 units. Each minor tick typically represents 1 or 2 units. U-100 is the global standard for insulin and peptide research dosing.

First calculate concentration (mg per mL) by dividing the total mg of peptide in the vial by the mL of bacteriostatic water you added. Convert to mcg per mL by multiplying by 1000. Then divide your desired dose in mcg by the mcg per mL to get volume in mL. Multiply that volume by 100 to get units on a U-100 syringe. This calculator does all of that automatically.

Bacteriostatic water is sterile water for injection containing 0.9 percent benzyl alcohol as a preservative, which inhibits bacterial growth in multi-use vials. The volume you use is flexible — more water gives a more dilute solution that is easier to draw accurately but means larger volumes per dose, while less water gives a more concentrated solution. Common reconstitution volumes are 1 to 5 mL depending on vial size and personal preference.

Concentration in mg per mL is the total mg of peptide in the vial divided by the volume of bacteriostatic water in mL. For example, 10 mg of peptide reconstituted with 2 mL of water gives a concentration of 5 mg per mL, or 5000 mcg per mL.

Divide the total amount of peptide in the vial by your per-dose amount. For example, a 10 mg vial dosed at 250 mcg (0.25 mg) gives 40 doses (10 divided by 0.25). This calculator displays doses per vial automatically and the cycle mode projects how many vials you need for a full cycle.
